Methotrexate Mechanism of Action – How Does Methotrexate Work?
The antimetabolite, methotrexate exerts its mechanism of action by preventing the metabolism the folic acid required for cell growth. Body cells require folic acid for DNA production. By inhibiting folic acid, Methotrexate hinders the cell's ability to repair and replicate its DNA.
This affects the growth of cells. Although Methotrexate targets normal body cells as well as cancer cells, it has more affinity towards fast growing cells such as tumor cells and bone marrow cells. The correct mechanism of action of Methotrexate to reduce inflammation (pain, swelling, stiffness) in rheumatoid arthritis is still unknown.
