Uses of Atavir (Atazanavir Sulfate Capsules)

Atavir (Atazanavir Sulfate capsules) are indicated in combination with other antiretroviral agents for the treatment of HIV-1 infection in adults and in pediatric patients 6 years of age and older weighing at least 15 kg.
Atazanavir will not cure or prevent HIV infection or AIDS. It helps keep HIV from reproducing and appears to slow down the destruction of the immune system. This may help delay problems that are usually related to AIDS or HIV disease from occurring. Atazanavir will not keep you from spreading HIV to other people. People who receive this medicine may continue to have other problems usually related to AIDS or HIV disease.

Atavir (Atazanavir Sulfate Capsules) - Dosage

Generic Atazanavir Sulfate capsules should be administered exactly as suggested by your doctor. Your doctor may recommend a dosage of Atavir capsules depending on your age, medical requirement, body weight, other medical conditions, and current medications.
Adults:
The recommended dose of Atavir capsules is 300 mg once daily taken with Ritonavir 100 mg once daily and with food. Ritonavir is used as a booster of Atazanavir pharmacokinetics.
Paediatric patients (6 years to less than 18 years of age and weighing at least 15 kg)
The dose of atazanavir capsules for paediatric patients is based on body weight and should not exceed the recommended adult dose. Atavir capsules must be taken with ritonavir and have to be taken with food.
Atavir capsules work effectively if the amount of Atazanavir Sulfate is maintained at a constant level in the body. You should be systematic about the daily schedule for the medication such that you take Atavir tablets at about the same time every day, to get the maximum benefit of the medication. Continue taking Atavir capsules without missing any doses for the recommended treatment period and do not stop the treatment suddenly even if you feel better.

Missed Dose

Atazanavir Sulfate Mechanism of Action

Atazanavir sulfate is an HIV-1 protease inhibitor that works by blocking the active site of the HIV-1 protease enzyme. This enzyme is crucial for the maturation of the virus, as it cleaves viral polyproteins into functional proteins needed for viral replication. By inhibiting this enzyme, Atazanavir prevents the formation of mature, infectious viral particles.

Storage Instructions for Atavir Capsules

Store Atavir capsules at controlled room temperature (20°C to 25°C),protected from heat, moisture, and sunlight. Excursions are permitted from 15°C to 30°C.
Retain the original packing of the medication and do not consume past its expiration date printed on the blister pack. Keep the pack of Atavir capsules away from the reach of children and pets. Do not dispose along with domestic waste or sewage. Consult your doctor or pharmacist about the correct method of disposal.

Contraindications

Atavir capsules are contraindicated in patients with a hypersensitivity (eg, Stevens-Johnson syndrome, erythema multiforme, or toxic skin eruptions) to Atazanavir Sulfate or any inactive ingredient present in Atavir capsules. Atavir capsules are contraindicated in the following cases:
Coadministration with Alfuzosin, Triazolam, orally administered midazolam, ergot derivatives, Rifampin, Irinotecan, Lovastatin, Simvastatin, Indinavir, Cisapride, Pimozide, St. John's wort, and Sildenafil.

Atazanavir (Atavir Capsules) Overdose

Human experience of acute overdose with Atazanavir is limited. Single doses up to 1200 mg have been taken by healthy volunteers without symptomatic untoward effects. A single self-administered overdose of 29.2 g of Atazanavir in an HIV-infected patient (73 times the 400-mg recommended dose) was associated with asymptomatic bifascicular block and PR interval prolongation. These events resolved spontaneously. At high doses that lead to high drug exposures, jaundice due to indirect (unconjugated) hyperbilirubinemia (without associated liver function test changes) or PR interval prolongation may be observed.
Treatment of overdosage with Atazanavir should consist of general supportive measures, including monitoring of vital signs and ECG, and observations of the patient's clinical status. If indicated, elimination of unabsorbed Atazanavir should be achieved by emesis or gastric lavage. Administration of activated charcoal may also be used to aid removal of unabsorbed drug. There is no specific antidote for overdose with Atazanavir (Atavir capsules).

Warnings and Precautions

Cardiac conduction abnormalities:
PR interval prolongation may occur in some patients. Use with caution in patients with preexisting conduction system disease or when administered with other drugs that may prolong the PR interval.
Rash:
Discontinue if severe rash develops.
Hyperbilirubinemia:
Most patients experience asymptomatic increases in indirect bilirubin, which is reversible upon discontinuation. Do not dose reduce. If a concomitant transaminase increase occurs, evaluate for alternative etiologies.
Hepatotoxicity:
Atavir Capsules should be used with caution in patients with hepatic impairment. Patients with hepatitis B or C infection are at risk of increased transaminases or hepatic decompensation. Monitor hepatic laboratory tests prior to therapy and during treatment.
Nephrolithiasis:
Nephrolithiasis has been reported. Consider temporary interruption or discontinuation.
Exacerbations Of Diabetes Mellitus/Hyperglycemia:
Patients receiving Atavir Capsules may develop new onset or exacerbations of diabetes mellitus/hyperglycemia, immune reconstitution syndrome, and redistribution/accumulation of body fat.
Hemophilia:
Spontaneous bleeding may occur and additional factor VIII may be required.

Side Effects of Atazanavir Sulfate (Atavir Capsules)

The most common Atazanavir side effects with an incidence rate of greater than 5% were asthenia, malaise, anorexia, nausea, paresthesia, circumoral paresthesia, peripheral paresthesia, jaundice/scleral icterus, rash, headache, abdominal pain, vomiting, insomnia, peripheral neurologic symptoms, dizziness, myalgia, diarrhea, depression, fever, and taste perversion.

Atazanavir Sulfate (Atavir Capsules) During Pregnancy

Atazanavir has been classified by the US FDA as Pregnancy Category B.
Atazanavir has been evaluated in a limited number of women during pregnancy. Available human and animal data suggest that Atazanavir does not increase the risk of major birth defects overall compared to the background rate.
Do not use this medicine without your doctor's consent in case you are pregnant. Stop using this medication and inform your doctor immediately if you become pregnant during treatment.

Nursing Considerations

HIV-1-infected mothers should not breastfeed their infants to avoid risking postnatal transmission of HIV-1 infection. Because of the potential for HIV-1 transmission mothers should be instructed not to breastfeed.
Do not breast-feed a baby if you are using Atavir Capsules.
