Valif Oral Jelly Mechanism of Action – How Does Vardenafil Hydrochloride Work?

Vardenafil belongs to class of drugs called phosphodiesterase type 5 inhibitor. Valif oral jelly prevents the activity of an enzyme called phosphodiesterase type 5.
An erection during sexual stimulation develops from a cascade of events. This process requires the signals from the nervous system which would result in the release of chemical messengers in the penis. One such chemical messenger involved in this process is called cyclic GMP. Cyclic GMP relaxes the blood vessels, resulting in an increase in the size of the blood vessels in the penis. This increase in blood vessel size permits more blood to flow into the penis, which ultimately results in the penis becoming firm and erect.
Usually, cyclic GMP is suppressed by another enzyme in the body called phosphodiesterase type 5. Valif oral jelly prevents the activity of phosphodiesterase type 5, thus inhibiting the suppression of cyclic GMP. If the cyclic GMPs are available to increase the size of blood vessels, the blood vessels are kept widened for a longer period of time, continuing the enhanced blood flow to the penis and maintaining a longer erection.
As Valif oral jelly indirectly boosts the activity of cyclic GMPS which are responsible for generating an erection, Vardenafil jelly will only have an effect once these messengers are in circulation i.e. sexual stimulation is required for cyclic GMPs to produce and sustain an erection. Contraindications

Valif Oral Jelly is contraindicated in patients with a hypersensitivity to Vardenafil or any inactive ingredient present in tablets. Administration of Vardenafil (Valif Oral Jelly) with nitrates (either regularly and/or intermittently) and nitric oxide donors is contraindicated.Consistent with the effects of PDE5 inhibition on the nitric oxide/cyclic guanosine monophosphate pathway, PDE5 inhibitors, including Vardenafil, may potentiate the hypotensive effects of nitrates.

Overdose

The maximum dose of Vardenafil (Valif Oral Jelly) for which human data are available is a single 120 mg dose administered to healthy male volunteers. The majority of these subjects experienced reversible back pain/myalgia and/or “abnormal vision.” Single doses up to 80 mg Vardenafil and multiple doses up to 40 mg Vardenafil administered once daily over 4 weeks were tolerated without producing serious adverse side effects. When 40 mg of Vardenafil was administered twice daily, cases of severe back pain were observed. No muscle or neurological toxicity was identified.
In cases of overdose, standard supportive measures should be taken as required. Renal dialysis is not expected to accelerate clearance as vardenafil is highly bound to plasma proteins and not significantly eliminated in the urine.

Medical History
