Uses of Penegra Tablets (Generic Sildenafil Citrate)

Penegra is a phosphodiesterase-5 (PDE5) inhibitor indicated for the treatment of erectile dysfunction (ED), also called sexual impotence. Sildenafil along with other medicines called called phosphodiesterase 5 (PDE5) inhibitors prevent an enzyme called phosphodiesterase type-5 from working too quickly. When a man is sexually stimulated, his body's normal response is to increase blood flow to his penis to produce an erection. By controlling the enzyme, Sildenafil helps to maintain an erection after the penis is stroked.
Sildenafil Citrate tablets are also used in both men and women to treat the symptoms of pulmonary arterial hypertension. This is a type of high blood pressure that occurs between the heart and the lungs. When hypertension occurs in the lungs, the heart must work harder to pump enough blood through the lungs. Sildenafil works on the PDE5 enzyme in the lungs to relax the blood vessels.

Penegra Tablets - Dosage

Generic Sildenafil Citrate Tablets tablets should be administered exactly as suggested by your doctor. Your doctor may recommend a dosage of Sildenafil tablets depending on your age, medical requirement, body weight, other medical conditions, and current medications.
Treatment of Erectile Dysfunction (ED):
For most patients, the recommended dose is 50 mg taken, as needed, approximately 1 hour before sexual activity. However, VIAGRA may be taken anywhere from 30 minutes to 4 hours before sexual activity

Contraindications

Penegra tablets are contraindicated in patients with a hypersensitivity to Sildenafil Citrate or any inactive ingredient present in tablets. Penegra tablets are also contraindicated in the following cases:

• Administration of Penegra to patients using nitric oxide donors, such as organic nitrates or organic nitrites in any form. VIAGRA was shown to potentiate the hypotensive effect of nitrates
• Administration with guanylate cyclase (GC) stimulators, such as Riociguat

Overdose

In studies with healthy volunteers of single doses of Sildenafil tablets up to 800 mg, adverse reactions were similar to those seen at lower doses but incidence rates and severities were increased. In cases of overdose, standard supportive measures should be adopted as required. Renal dialysis is not expected to accelerate clearance as Sildenafil is highly bound to plasma proteins and it is not eliminated in the urine.

Warnings and Precautions

• Patients should not use Penegra if sexual activity is inadvisable due to cardiovascular status
• Patients should seek emergency treatment if an erection lasts for more than 4 hours. Use Penegra with caution in patients predisposed to priapism
• Patients should stop Penegra and seek medical care if a sudden loss of vision occurs in one or both eyes, which could be a sign of non arteritic anterior ischemic optic neuropathy (NAION). Penegra should be used with caution, and only when the anticipated benefits outweigh the risks, in patients with a history of NAION. Patients with a "crowded" optic disc may also be at an increased risk of NAION
• Patients should stop Penegra and seek prompt medical attention in the event of sudden decrease or loss of hearing
• Caution is advised when Penegra is co-administered with alpha-blockers or anti-hypertensives. Concomitant use may lead to hypotension
• Decreased blood pressure, syncope, and prolonged erection may occur at higher sildenafil exposures. In patients taking strong CYP inhibitors, such as Ritonavir, Sildenafil exposure is increased. Decrease in Penegra dosage is recommended.
• Consumption of excess alcohol can obstruct your capacity to achieve an erection. The effect of Sildenafil tablets (Penegra tablets) may be lost in such a scenario. Alcohol ingestion can also increase the possibility of certain side effects.

Side Effects of Penegra Tablets (Sildenafil Citrate Tablets)

Side effects reported by 2% or more of patients treated with Sildenafil and more frequent than placebo during clinical trials include Headache, Flushing, Dyspepsia, Abnormal vision*, Nasal congestion, Back pain, Myalgia, Nausea, Dizziness and Rash. • Hemic and Lymphatic: anemia and leukopenia.
• Metabolic and Nutritional: thirst, edema, gout, unstable diabetes, hyperglycemia, peripheral edema, hyperuricemia, hypoglycemic reaction, hypernatremia.
• Musculoskeletal: arthritis, arthrosis, myalgia, tendon rupture, tenosynovitis, bone pain, myasthenia, synovitis.
• Nervous:ataxia, hypertonia, neuralgia, neuropathy, paresthesia, tremor, vertigo, depression, insomnia, somnolence, abnormal dreams, reflexes decreased, hypesthesia.

• Special Senses: sudden decrease or loss of hearing, mydriasis, conjunctivitis, photophobia, tinnitus, eye pain, ear pain, eye hemorrhage, cataract, dry eyes.
• Urogenital: cystitis, nocturia, urinary frequency, breast enlargement, urinary incontinence, abnormal ejaculation, genital edema and anorgasmia.

Sildenafil Citrate (Penegra Tablets ) During Pregnancy

Sildenafil Citrate has been classified by the US FDA as Pregnancy Category B.
Limited published data from randomized controlled trials, case-controlled trials, and case series do not report a clear association with sildenafil and major birth defects, miscarriage, or adverse maternal or fetal outcomes when Sildenafil is used during pregnancy (for the treatment of Pulmonary Arterial Hypertension).
No evidence of teratogenicity, embryotoxicity, or fetotoxicity was observed in pregnant rats or rabbits dosed with sildenafil 200 mg/kg/day during organogenesis, a level that is, on a mg/m basis, 32- and 65-times, respectively, the recommended human dose (RHD) of 20 mg three times a day.
Do not use this medicine without your doctor's consent in case you are pregnant. Stop using this medication and inform your doctor immediately if you become pregnant during treatment.
Limited published data from a case report describe the presence of sildenafil and its active metabolite in human milk. There is insufficient information about the effects of Sildenafil on the breastfed infant and no information on the effects of Sildenafil on milk production. Limited clinical data during lactation preclude a clear determination of the risk of Sildenafil to an infant during lactation.
