Norethisterone Tablets (Primolut N) - Product Information

Norethisterone tablets belong to a class of medications known as progestogens, which imitate the mechanism of the naturally occurring female hormone called progesterone. Norethisterone tablets are used for the prevention of unusual or untimely uterus bleeding, reducing symptoms of pre-menstrual syndrome, and easing painful or heavy periods (menorrhagia).
Primolut N tablets (Norethisterone) are also used for the treatment of endometriosis, primary and secondary amenorrhoea (lack of monthly periods), certain types of breast cancer, and for the postponement of menstrual periods. Norethisterone tablets are also used as contraception to avoid pregnancies. Norethisterone tablets are available in strength of 5 mg.

Each Primolut N tablet contains 5 mg of Norethisterone as the active pharmaceutical ingredient. The inactive ingredients in Primolut N tablets are lactose, maize starch, and magnesium stearate.

What are Primolut N Tablets Used For?

Norethisterone is a synthetic version of the naturally produced female sex hormone called progesterone. Norethisterone belongs to a class of medication known as progestogens because they produce the same effects as the female progesterone hormone. Primolut N tablets are used for the treatment of erratic, painful or excessive blood flow during menstrual periods. Norethisterone tablets are also beneficial for the treatment of endometriosis and certain cases of breast cancer.
Norethisterone tablets alleviate the symptoms of pre-menstrual syndrome such as breast pain, headache, migraine, fluid retention, and mood swings. Primolut N tablets are also used to delay the regular monthly menstrual cycle by a few days.
Norethisterone is also used as a contraceptive. It is popularly known as a progestogen-only oral contraceptive or the mini pill. Norethisterone tablets thicken the vaginal mucus at the cervix which prevents the entry of the sperm into the uterus. It also modifies the endometrial lining of the womb which inhibits the attachment of a fertilized egg to the uterus. Using these mechanisms, Norethisterone tablets effectively avert the incidence of pregnancy in women. Drug Dosage- How to take Norethisterone tablets 

Primolut N tablets are prescribed for different medical conditions. Your dosage and treatment period will vary depending on your medical condition and your response to treatment. Take Norethisterone tablets exactly as prescribed by your doctor. You can consume Norethisterone tablets before or after meals. Norethisterone tablets should be administered along with a full glass of water. Try to maintain a regular schedule for the medication such that you take the medication at the same time every day.
Consult your doctor about using Norethisterone tablets for contraception purposes. On completing a full course of Norethisterone tablets as birth control pills, your body will go through a withdrawal bleed. If this bleeding does not occur, please consult your doctor because you should make sure that you aren’t pregnant before starting a new pack. Given below are dosing schedules for different medical conditions:
Primolut N to Stop Abnormal bleeding – 1 Norethisterone tablet three times a day for a period of 10 days. Bleeding usually stops within 1-3 days. A menstrual period usually begins 2-4 days after the treatment period.
Prevention of Abnormal Bleeding – 1 Norethisterone tablet twice a day from the 19th - 26th day of the cycle.
Premenstrual Syndrome - 2 to 3 Norethisterone tablets a day from the 19th - 26th day of the cycle. This treatment may continue for some months.
Norethisterone Dosage to Delay a period - 1 Norethisterone tablet three times a day starting three days before the expected beginning of the period. Your period should occur 2-3 days after you have stopped taking the medicine.
Painful periods - 1 Norethisterone tablet three times a day for 20 days starting on the 5th day of the cycle. You will usually be treated for 3-4 cycles.
Norethisterone Tablets for Heavy periods - 1 Norethisterone tablet two to three times a day from the 19th - 26th day of the cycle.
Endometriosis - 2 Norethisterone tablets a day starting on the 5th day of your cycle. If you continue to experience spotting, the dosage may
be increased to 4 to 5 tablets a day. Once bleeding has stopped the dosage may be reduced. Your treatment may last for about 4-6 months or longer.
Breast Cancer - 8 Norethisterone tablets a day, the dosage may be increased to 12 tablets a day.

Missed Dose

If you miss your regular dose of Norethisterone tablets, take it as soon as you remember. You can continue you regular schedule the next day. If you remember the missed Norethisterone dose the next day, take your regular dose and skip the missed dose. Do not take a double dose to compensate the missed dose.
If you have been prescribed Norethisterone tablets for contraception purposes and have missed a dose, you can take the missed dose as soon as you remember. However, if more than 3 hours have passed after your scheduled time, you will have to practice an additional non-hormonal method of contraception (condoms) for the next 2 days.

Primolut N Mechanism of Action – How Does Norethisterone Work?

Norethisterone has similar outcomes to those of the naturally occurring female sex hormone, progestogen. Norethisterone alters the endometrium lining from the proliferative phase to the secretory phase. This feature is useful for the treatment of excessive menstrual bleeding, endometriosis, and postponement of menstrual periods. Norethisterone tablets may also exhibit certain estrogenic, anabolic and androgenic functions.
Primolut N tablets suppress certain ovarian functions which result in the effective reduction of the medical conditions related to pre-menstrual syndrome. Norethisterone tablets are used as birth control pills because of its negative feedback inhibition of the pituitary gland hormone called gonadotropin, resulting in the prevention of ovulation.

Storage Instructions for Norethisterone Tablets

Store Norethisterone tablets at controlled room temperature (20°C to 25°C), protected from heat, sunlight, and moisture. Excursions are permitted from 15°C to 30°C.
Keep Norethisterone tablets in its original blister pack. Do not consume Primolut N tablets after the expiration date printed on the blister pack has passed. Make sure that the pack of Primolut N tablets are away from the reach of children and pets. Do not dispose Norethisterone tablets along with other domestic waste or sewage. Consult your pharmacist for additional details about the correct disposal technique.

Contraindications

Primolut N tablets are contraindicated in patients who have a known hypersensitivity to the active ingredient Norethisterone or to any of the inactive substances present in Primolut N tablets.
They are also contraindicated in cases of known or suspected pregnancy, breast-feeding, thromboembolic disorders, thrombophlebitis, cerebral apoplexy or a history of these conditions, arterial and cardiovascular disease (myocardial infarction, cerebrovascular accident, ischemic heart disease) present in the patient or history of the same, diabetes mellitus with vascular involvement, presence or history of severe hepatic disease as long as liver function values have not returned to normal, presence or history of liver tumours (benign or malignant), known or suspected sex hormone-dependent malignancies, Dubin-Johnson syndrome, Rotor syndrome, abortion, undiagnosed vaginal or urinary bleeding, and undiagnosed breast pathology.

Overdose

There have been no reports of ill-effects from overdosage and treatment is generally unnecessary. There are no special antidotes, and treatment should be symptomatic.

Warnings and Precautions to be taken when using Norethisterone Tablets

Medical History
Before starting the course of Norethisterone tablets, let your doctor known about your personal and family medical history, including any present medical conditions that you may have recently developed. Primolut N tablets should not be consumed by patients who have any liver disease, jaundice or herpes during pregnancy, Dubin-Johnson Syndrome (chronic jaundice (yellowing of the skin or eyes)) or Rotor Syndrome (jaundice in childhood), irregular vaginal bleeding of unknown cause, blood clot affecting the legs (deep vein thrombosis) or lungs (pulmonary embolism), recent heart attack (myocardial infarction) or angina, an inherited disorder of the red blood pigment haemoglobin (porphyria), severe itching, diabetes, epilepsy, migraine, asthma, kidney heart problems, depression, and cancer of the breast or genital tract (unless Norethisterone tablets are given as part of a cancer therapy). Consult your doctor before taking Norethisterone tablets if you have missed your monthly menstrual period before beginning a new course of Norethisterone tablets because pregnant women should not consume Norethisterone tablets. Breast-feeding women should also inform their doctor before consuming Norethisterone tablets.
Allergy

Drug Interactions
Before using Primolut N tablets, please let your doctor know about all the medications you are using currently such as prescription medications, over the counter drugs, vitamins, mineral supplements, and herbal remedies. Inform your doctor if you are using or have used these drugs in the recent past: hormonal products (oestrogens or progestogens), epilepsy medications (phenobarbital, phenytoin, carbamazepine), antibiotics (rifampicin, nevirapine, tetracyclines, ampicillin, oxacillin, cotrimoxazole, ritonavir, nelfinavir), St John's Wort (hypericum perforatum-a herbal remedy), aminoglutethimide, cyclosporin (to prevent organ transplant rejection), anti-inflammatory medicines (e.g. ibuprofen) or vasodilators.
Driving and Using Machines
Norethisterone tablets may induce dizziness, drowsiness or impair your vision. Ensure that your mental stability, alertness, and attention span are not affected by the administration of Norethisterone tablets before you perform activities such as driving or operating machinery.
Sugar Intolerance
Lactose is among the inactive ingredients present in Norethisterone tablets. If you have a known intolerance to some sugars, consult your doctor before starting the course of Norethisterone tablets.
Tests

  • Skin: acne, hair growth (especially on the top lip or chin), hair loss, hirsutism, itchiness, worsening of existing skin conditions, sweating
  • Hypersensitivity reactions: severe allergic reaction including skin rash, hives, swelling of the face, lips, tongue or throat, or difficulty breathing or swallowing
  • Other: fluid retention, bloating, high fever, change in appetite, deepening of the voice, changes in the sex drive, Cushing's syndrome,
