Nitazoxanide is an antiprotozoal agent used to treat diarrhea caused by Cryptosporidium parvum or Giardia lamblia in patients 12 years and older.
Nitazoxanide tablets have not been shown to be effective for the treatment of diarrhea caused by Cryptosporidium parvum in HIV-infected or immunodeficient patients.

Uses of Nitazoxanide Tablets (Nizonide Tablets)
Nitazoxanide is an antiprotozoal medicine that treats infections caused by protozoa (single-cell parasites that live in moist places such as lakes, streams, and soil).
Nitazoxanide is used to treat diarrhea caused by Giardia or Cryptosporidium. These conditions are sometimes called "Traveler's diarrhea." Nitazoxanide tablets are used in adults and children who are at least 12 years old. Nitazoxanide oral suspension is indicated for patients at least 1 year of age. Diarrhea is loose or watery stool.
Nizonide Tablets - Dosage
Generic Nitazoxanide tablets should be administered exactly as suggested by your doctor. Your doctor may recommend a dosage of Nizonide tablets depending on your age, medical requirement, body weight, other medical conditions, and current medications.
Nitazoxanide tablets should not be administered to pediatric patients 11 years of age or younger because a single tablet contains a greater amount of nitazoxanide than the recommended dosing in this pediatric age group.
The recommended dosage for patients 12 years and older is One Nitazoxanide tablet (500 mg nitazoxanide) taken orally every 12 hours with food. The duration of treatment is 3 days.
Usual Adult Dose for Cryptosporidiosis:

Nizonide tablets work effectively if the amount of Nitazoxanide is maintained at a constant level in the body. You should be systematic about the daily schedule for the medication such that you take tablets at about the same time every day, to get the maximum benefit of the medication. Continue taking Nitazoxanide tablets without missing any doses for the recommended treatment period and do not stop the treatment suddenly even if you feel better.

Missed Dose
If you forget to take your regular Nizonide tablet dose on time, take it whenever you remember.
If you remember the next day, skip the missed dose and take your scheduled dose. Do not take a double dose to compensate the missed dose.
Mechanism of Action
Energy Disruption:
Once administered, Nitazoxanide is rapidly metabolized into its active form, tizoxanide.
PFOR Inhibition:
Tizoxanide interferes with the pyruvate:ferredoxin oxidoreductase (PFOR) enzyme-dependent electron transfer reaction, which is essential for anaerobic energy metabolism in susceptible parasites and anaerobic bacteria.
Antiviral Activity (Investigational):
While primarily used as an antiprotozoal, the drug is also researched for antiviral effects. It is believed to inhibit viral replication by disrupting the maturation of viral glycoproteins during the post-translational stage, thereby preventing host cell entry and assembly
Storage Instructions for Nizonide Tablets
Store Nizonide tablets at controlled room temperature (20°C to 25°C),protected from heat, moisture, and sunlight. Excursions are permitted from 15°C to 30°C.
Retain the original packing of the medication and do not consume past its expiration date printed on the blister pack. Keep the pack of Nizonide tablets away from the reach of children and pets. Do not dispose Nitazoxanide tablets along with domestic waste or sewage. Consult your doctor or pharmacist about the correct method of disposal.
Contraindications
Nizonide tablets are contraindicated in patients with a hypersensitivity to Nitazoxanide or any inactive ingredient present in Nizonide tablets.
Overdose
Limited information on nitazoxanide overdosage is available. In the event of overdose, gastric lavage may be appropriate soon after oral administration. Patients should be observed and given symptomatic and supportive treatment.
There is no specific antidote for overdose with Nitazoxanide. Tizoxanide is the primary active metabolite of Nitazoxanide. Because Tizoxanide is highly protein bound, dialysis is unlikely to significantly reduce plasma concentrations of the drug.
Warnings and Precautions
Patients With Hepatic, Renal And Biliary Disease:
Nitazoxanide should be administered with caution to patients with hepatic, renal and biliary disease.
Pre-Existing Health Conditions:
Discuss any pre-existing health conditions with their doctor. Undergo liver function tests if there is a history of liver disease.

It is very important that your doctor check your progress at regular visits and to check for unwanted effects.If your symptoms do not improve within a few days or if they become worse, check with your doctor.
Highly Protein Bound Drugs with Narrow Therapeutic Indices:
Tizoxanide (the active metabolite of Nitazoxanide) is highly bound to plasma protein. Therefore, monitor for adverse reactions when administering Nitazoxanide concurrently with other highly plasma protein-bound drugs with narrow therapeutic indices, as competition for binding sites may occur (e.g., warfarin).
Side Effects of Nizonide Tablets
The most frequent side effects with an incidence rate of 2% or more include abdominal pain, headache, nausea, and chromaturia (a benign discoloration of the urine).
These symptoms are usually mild to moderate in severity and tend to resolve on their own. Some patients might experience headaches, dizziness, or a slightly elevated liver enzyme level, which typically normalizes after discontinuation of the drug. More serious but rare side effects can include hypersensitivity reactions such as rashes or pruritus, and in extremely rare cases, severe allergic reactions like anaphylaxis.
Nitazoxanide Tablets During Pregnancy
Nitazoxanide tablets are classified by the US FDA as Pregnancy Category B.
There are no data with Nitazoxanide in pregnant women to inform a drug-associated risk. No teratogenicity or fetotoxicity was observed in animal reproduction studies with administration of nitazoxanide to pregnant rats and rabbits during organogenesis at exposures 30 and 2 times, respectively, the exposure at the maximum recommended human dose of 500 mg twice daily based on body surface area (BSA).

Nursing Considerations
No information regarding the presence of Nitazoxanide in human milk, the effects on the breastfed infant, or the effects on milk production is available.
