Uses of Flibanserin Tablets (Fliban)

Flibanserin is used to treat low sexual desire in women who have not gone through menopause and have never had low sexual desire in the past. Flibanserin is for use only when low sexual desire is NOT caused by a medical condition, a mental disorder, relationship problems, or by using drugs or other medicines.
Flibanserin is not for women who have already gone through menopause. flibanserin is also not for use by men.

Flibanserin Tablets (Fliban) - Dosage

Generic Flibanserin tablets should be administered exactly as suggested by your doctor. Your doctor may recommend a dosage of Fliban tablets depending on your age, medical requirement, body weight, other medical conditions, and current medications.
The recommended dosage is 100 mg taken once daily at bedtime. Fliban is dosed at bedtime because administration during waking hours increases risks of hypotension, syncope, accidental injury, and central nervous system (CNS) depression.
Discontinue treatment with Fliban tablets after 8 weeks if no improvement.

Mechanism of Action

Flibanserin's mechanism of action involves modulating neurotransmitters in the brain to treat hypoactive sexual desire disorder (HSDD) by increasing sexual desire. It selectively targets serotonin receptors in the prefrontal cortex, acting as an agonist on 5- HT-1A receptors and an antagonist on 5- HT-2A receptors.

Contraindications

Overdosage of Flibanserin may cause an increase in the incidence or severity of any of the reported adverse reactions. In the event of overdosage, treatment should address the symptoms and supportive measures, as needed. There is no known specific antidote for Fliban Tablets (Generic Flibanserin).

Warnings and Precautions

Hypotension and Syncope with Flibanserin Alone:
Patients with pre-syncope should immediately lie supine and promptly seek medical help if symptoms do not resolve.
Central Nervous System (CNS) Depression (e.g., Somnolence, Sedation):
Can occur with Flibanserin alone. Exacerbated by other CNS depressants, and in settings where flibanserin concentrations are increased. Patients should avoid activities requiring full alertness (e.g., operating machinery or driving) until at least 6 hours after each dose and until they know how Flibanserin affects them
Hypotension and Syncope due to an Interaction with Alcohol
Taking Flibanserin within two hours after consuming alcohol increases the risk of severe hypotension and syncope. To reduce this risk, counsel patients to wait at least two hours after drinking one or two standard alcoholic drinks before taking Flibanserin at bedtime
Hypotension and Syncope with CYP3A4 Inhibitors
Moderate or Strong CYP3A4 Inhibitors:The concomitant use of Flibanserin with moderate or strong CYP3A4 inhibitors significantly increases flibanserin concentrations, which can lead to hypotension and syncope. The concomitant use of Flibanserin with a moderate or strong CYP3A4 inhibitor is contraindicated.

Syncope and Hypotension in Patients with Hepatic Impairment:
The use of Flibanserin in patients with any degree of hepatic impairment significantly increases flibanserin concentrations, which can lead to hypotension and syncope. Therefore, the use of Flibanserin is contraindicated in patients with hepatic impairment.
Hypersensitivity Reactions:
Hypersensitivity reactions, including anaphylaxis, reactions consistent with angioedema (e.g., swelling of the face, lips, and mouth), pruritus, and urticaria have been reported with Flibanserin. Flibanserin is contraindicated in women with known hypersensitivity to Flibanserin or any of its components.

Side Effects of Flibanserin Tablets (Fliban)

Most common side efffects with an incidence rate of 2% or greater are dizziness, somnolence, nausea, fatigue, insomnia, and dry mouth.
In four trials in premenopausal women with HSDD treated with 100 mg Flibanserin at bedtime, less common side effects (reported in 1% or more but <2% of Flibanserin-treated patients and at a higher incidence than with placebo) included:

• Anxiety (Flibanserin 1.8%; placebo 1.0%)
• Constipation (Flibanserin 1.6%; placebo 0.4%)
• Abdominal pain (Flibanserin 1.5%; placebo 0.9%)
• Metrorrhagia (Flibanserin 1.4%; placebo 1.4%)
• Rash (Flibanserin 1.3%; placebo 0.8%)
• Sedation (Flibanserin 1.3%; placebo 0.2%)
• Vertigo (Flibanserin 1%; placebo 0.3%)

Flibanserin During Pregnancy

Flibanserin (Fliban Tablets) has been classified by the US FDA as Pregnancy Category Not Assigned.
There are no studies of Flibanserin in pregnant women to inform whether there is a drug-associated risk in humans. In animals, fetal toxicity only occurred in the presence of significant maternal toxicity including reductions in weight gain and sedation. Adverse reproductive and developmental effects consisted of decreased fetal weight, structural anomalies and increases in fetal loss at exposures greater than 15 times exposures achieved with the recommended human dosage.
