Finalo Tablets Mechanism of Action – How Does Finasteride Work?

Finasteride is a competitive and specific inhibitor of type II 5a -reductase. Finasteride has no affinity for the androgen receptor and has no androgenic, anti-androgenic, oestrogenic, anti-oestrogenic, or progestational effects. Inhibition of this enzyme blocks the peripheral conversion of testosterone to the androgen DHT, resulting in significant decreases in serum and tissue DHT concentrations. Finasteride produces a rapid reduction in serum DHT concentration, reaching significant suppression within 24 hours of dosing.
Hair follicles contain type II 5a -reductase. In men with male pattern hair loss, the balding scalp contains miniaturised hair follicles and increased amounts of DHT. Administration of finasteride decreases scalp and serum DHT concentrations in these men. Men with a genetic deficiency of type II 5a -reductase do not suffer from male pattern hair loss. Finasteride inhibits a process responsible for miniaturisation of the scalp hair follicles, which can lead to reversal of the balding process.

Storage

Store Finalo 1 mg tablets at controlled room temperature (20°C to 25°C),protected from heat, moisture, and sunlight. Excursions are permitted from 15°C to 30°C.
Do not consume Finalo tablets after its expiration date printed on the blister pack. Do not dispose generic Finasteride 1 mg tablets along with other domestic waste. Finalo tablets should be discarded in an environment-friendly manner.

Finalo Tablets - Contraindications

Finalo tablets are contraindicated in patients with a hypersensitivity to Finasteride or any inactive ingredient present in tablets. Finalo tablets are also contraindicated in the following cases:
Pregnancy. Finasteride use is contraindicated in women when they are or may potentially be pregnant. Because of the ability of Type II 5 a-reductase inhibitors to inhibit the conversion of testosterone to 5a-dihydrotestosterone (DHT), finasteride may cause abnormalities of the external genitalia of a male fetus of a pregnant woman who receives finasteride. If this drug is used during pregnancy, or if pregnancy occurs while taking this drug, the pregnant woman should be apprised of the potential hazard to the male fetus.
Finalo Tablets should not be taken by men who are taking Finasteride 5 mg or any other 5 alpha-reductase inhibitor for benign prostatic hyperplasia or any other condition.

Overdose

In clinical studies, single doses of Finasteride (Finalo 1 mg tablets) up to 400 mg and multiple doses of Finasteride up to 80 mg/day for three months did not result in adverse reactions. Until further experience is obtained, no specific treatment for an overdose with Finasteride can be recommended.
Significant lethality was observed in male and female mice at single oral doses of 1500 mg/m2 (500 mg/kg) and in female and male rats at single oral doses of 2360 mg/m2 (400 mg/kg) and 5900 mg/m2 (1000 mg/kg), respectively.

Warnings and Precautions

Other causes of alopecia should be ruled out prior to prescribing Finasteride (Finalo 1 mg tablets). Efficacy and duration of treatment should be assessed periodically by the treating physician.
Paediatric population
Finalo 1 mg tablets should not be used in women or pediatric patients. There are no data demonstrating efficacy or safety of Finasteride in children under the age of 18.
Pregant Women
Women should not handle crushed or broken Finalo tablets when they are pregnant or may potentially be pregnant due to potential risk to a male fetus
Effects on Prostate Specific Antigen (PSA)
In clinical studies with 'Finalo' in men 18-41 years of age, the mean value of serum prostate-specific antigen (PSA) decreased from 0.7 ng/ml at baseline to 0.5 ng/ml at month 12. Doubling the PSA level in men taking 'Finalo' should be considered before evaluating this test result. Any confirmed increase in PSA while on Finalo may signal the presence of prostate cancer and should be evaluated, even if those values are still within the normal range for men not taking a 5alpha-reductase inhibitor.
Hepatic Impairment
The effect of hepatic insufficiency on the pharmacokinetics of Finasteride has not been studied. Caution should be used in the administration of Finasteride (Finalo 1 mg tablets) in patients with liver function abnormalities, as Finasteride is metabolized in the liver.
Breast Cancer
Breast cancer has been reported in men taking Finasteride 1 mg during the post-marketing period. Physicians should instruct their patients to promptly report any changes in their breasts such as lumps, pain or nipple discharge. Breast changes including breast enlargement, tenderness and neoplasm have been reported
Mood alterations and depression
Mood alterations including depressed mood, depression and, less frequently, suicidal ideation have been reported in patients treated with Finasteride 1 mg.
