The active ingredient present in D-Veniz tablets is Desvenlafaxine Succinate. Each tablet of D-Veniz contains Desvenlafaxine Succinate equivalent to 50 mg or 100 mg of Desvenlafaxine.

Uses of Desvenlafaxine Extended Release Tablets (D-Veniz)

D-Veniz (Desvenlafaxine Extended Release Tablets) is used to treat depression. It belongs to a group of medicines known as serotonin and norepinephrine reuptake inhibitors (SNRIs). These medicines are thought to work by increasing the activity of chemicals called serotonin and norepinephrine in the brain.
A major depressive episode (DSM-IV) implies a prominent and relatively persistent (nearly every day for at least 2 weeks) depressed or dysphoric mood that usually interferes with daily functioning, and includes at least 5 of the following 9 symptoms: depressed mood, loss of interest in usual activities, significant change in weight and/or appetite, insomnia or hypersomnia, psychomotor agitation or retardation, increased fatigue, feelings of guilt or worthlessness, slowed thinking or impaired concentration, or a suicide attempt or suicidal ideation.

D-Veniz Tablets (Generic Desvenlafaxine Extended Release Tablets) - Dosage

Generic Desvenlafaxine Extended Release tablets should be administered exactly as suggested by your doctor. Your doctor may recommend a dosage of D-Veniz tablets depending on your age, medical requirement, body weight, other medical conditions, and current medications.
The recommended dose is 50 mg once daily with or without food. There was no evidence that doses greater than 50 mg per day confer any additional benefit.
The 25 mg per day dose is intended for a gradual reduction in dose when discontinuing treatment or dosing in severe renal and end-stage renal disease patient.
D-Veniz tablets work effectively if the amount of Desvenlafaxine is maintained at a constant level in the body. You should be systematic about the daily schedule for the medication such that you take tablets at about the same time every day, to get the maximum benefit of the medication. Continue taking D-Veniz tablets without missing any doses for the recommended treatment period and do not stop the treatment suddenly even if you feel better.

Missed Dose

If you forget to take your regular D-Veniz tablet dose on time, take it whenever you remember.
If you remember the next day, skip the missed dose and take your scheduled dose. Do not take a double dose to compensate the missed dose.

Desvenlafaxine Succinate Mechanism of Action

The exact mechanism of the antidepressant action of Desvenlafaxine is unknown but is thought to be related to the potentiation of serotonin and norepinephrine in the central nervous system, through inhibition of their reuptake.

Storage Instructions for D-Veniz Tablets (Generic Desvenlafaxine Extended Release Tablets)

Store D-Veniz Tablets at controlled room temperature (20°C to 25°C),protected from heat, moisture, and sunlight. Excursions are permitted from 15°C to 30°C.
Retain the original packing of the medication and do not consume past its expiration date printed on the blister pack. Keep the pack of Generic Desvenlafaxine Extended Release Tablets away from the reach of children and pets. Do not dispose along with domestic waste or sewage. Consult your doctor or pharmacist about the correct method of disposal.

Contraindications

D-Veniz tablets are contraindicated in patients with a hypersensitivity to Desvenlafaxine Succinate, Venlafaxine Hydrochloride or any inactive ingredient present in tablets. D-Veniz tablets are contraindicated in the following cases:
Serotonin Syndrome and MAOIs: Do not use MAOIs intended to treat psychiatric disorders with D-Veniz tablets or within 7 days of stopping treatment with D-Veniz tablets. Do not use D-Veniz Tablets (Desvenlafaxine Extended Release Tablets) within 14 days of stopping an MAOI intended to treat psychiatric disorders. In addition, do not start Desvenlafaxine Extended Release tablets in a patient who is being treated with linezolid or intravenous methylene blue.

Overdose

In postmarketing experience, overdose with Venlafaxine (the parent drug of D-Veniz Tablets) has occurred predominantly in combination with alcohol and/or other drugs. The most commonly reported events in overdosage include tachycardia, changes in level of consciousness (ranging from somnolence to coma), mydriasis, seizures, and vomiting. Electrocardiogram changes (e.g., prolongation of QT interval, bundle branch block, QRS prolongation), sinus and ventricular tachycardia, bradycardia, hypotension, rhabdomyolysis, vertigo, liver necrosis, serotonin syndrome, and death have been reported.
No specific antidotes for Desvenlafaxine (D-Veniz tablets) are known. In managing over dosage, consider the possibility of multiple drug involvement.

Warnings and Precautions

Serotonin Syndrome:
Increased risk when co-administered with other serotonergic agents, but also when taken alone. If it occurs, discontinue Desvenlafaxine and serotonergic agents and initiate supportive treatment
Elevated Blood Pressure:
Control hypertension before initiating treatment. Monitor blood pressure regularly during treatment
Increased Risk of Bleeding:
Concomitant use of aspirin, NSAIDs, other antiplatelet drugs, warfarin, and other anticoagulants may increase this risk
Angle Closure Glaucoma:
Avoid use of antidepressants, including Desvenlafaxine, in patients with untreated anatomically narrow angles treated
Activation of Mania/Hypomania:
Use cautiously in patients with Bipolar Disorder. Caution patients about risk of activation of mania/hypomania
Discontinuation Syndrome:
Taper dose when possible and monitor for discontinuation symptoms
Seizure:
Can occur. Use cautiously in patients with seizure disorder
Hyponatremia:
Can occur in association with SIADH
Interstitial Lung Disease and Eosinophilic Pneumonia:
Can occur
Sexual Dysfunction:
Desvenlafaxine may cause symptoms of sexual dysfunction

Side Effects of Desvenlafaxine (D-Veniz Tablets)

Most common side effects with an incidence rate of 5% or greater and twice the rate of placebo in the 50 or 100 mg dose groups) were: nausea, dizziness, insomnia, hyperhidrosis, constipation, somnolence, decreased appetite, anxiety, and specific male sexual function disorder.
Other infrequent side effects, Observed in Premarketing and Postmarketing Clinical Studies , occurring at an incidence of less than 2% in Major Depressive Disorder (MDD) patients treated with desvenlafaxine extended-release tablets were:

  • Cardiac disorders - Tachycardia.
  • General disorders and administration site conditions - Asthenia.
  • Investigations - Weight increased, liver function test abnormal, blood prolactin increased.
  • Musculoskeletal and connective tissue disorders - Musculoskeletal stiffness.
  • Nervous system disorders - Syncope, convulsion, dystonia.
  • Psychiatric disorders - Depersonalization, bruxism.
  • Renal and urinary disorders - Urinary retention.
  • Skin and subcutaneous tissue disorders - Rash, alopecia, photosensitivity reaction, angioedema.

 

Desvenlafaxine (D-Veniz Tablets) During Pregnancy

Desvenlafaxine (D-Veniz Tablets) has been classified by the US FDA as Pregnancy Category .
There are no published studies on Desvenlafaxine in pregnant women; however published epidemiologic studies of pregnant women exposed to venlafaxine, the parent compound, have not reported a clear association with adverse developmental outcomes.
In reproductive developmental studies in rats and rabbits treated with desvenlafaxine succinate, there was no evidence of teratogenicity at a plasma exposure (AUC) that is up to 19-times (rats) and 0.5-times (rabbits) the exposure at an adult human dose of 100 mg per day. However, fetotoxicity and pup deaths were observed in rats at 4.5-times the AUC exposure observed with an adult human dose of 100 mg per day.
Do not use this medicine without your doctor's consent in case you are pregnant. Stop using this medication and inform your doctor immediately if you become pregnant during treatment.

Desvenlafaxine (D-Veniz Tablets) Nursing Considerations

Available limited data from published literature show low levels of Desvenlafaxine in human milk, and have not shown adverse reactions in breastfed infants (see Data). There are no data on the effects of desvenlafaxine on milk production.
The developmental and health benefits of breastfeeding should be considered along with the mother’s clinical need for Desvenlafaxine extended-release tablets and any potential adverse effects on the breastfed child from desvenlafaxine extended-release tablets or from the underlying maternal condition.
Do not use this medication without telling your doctor if you are breast-feeding a baby.
