Avana Mechanism of Action – How Does Avanafil Work?

Avanafil is an orally administered tablet, which is used for the treatment of male impotence (the inability to achieve or sustain a penile erection), also known as erectile dysfunction (ED). The half-life of Avanafil tablets in the human body is approximately 5 hours. Avanafil tablets belong to a class of medications known as phosphodiesterase type 5 inhibitors. This class of drugs also includes other erectile dysfunction medications like Tadalafil (Cialis), Sildenafil (Viagra) and Vardenafil (Levitra).
Erection of the penis is caused by the increased blood flow into the penis. This increased blood flow occurs due to the expansion of the size of the blood vessels that bring blood to the penis. Simultaneously, there is a dilation in the size of the blood vessels that take blood away from the penis so that there is lesser removal of blood from the penis. During sexual stimulation, penis erection results in the production and release of nitric oxide in the penis. This nitric oxide in turn causes an enzyme called guanylate cyclase to produce cyclic guanosine monophosphate (cGMP).
cGMP mainly controls the extension and dilation of the size of the blood vessels which transport blood to and from the penis during penile erection. However, the cGMP molecules are inhibited by a different enzyme called phosphodiesterase-5. Due to cGMP inhibition by phosphodiesterase-5 enzyme, the blood vessels shrink to their original size, resulting in lesser blood flow to the penis. Low blood flow causes the loss of penile erection.
Avanafil tablets prevent phosphodiesterase-5 enzyme from affecting cGMP molecules, so that cGMP can continue its activity of increasing the blood flow to the penis.
The presence of cGMP leads to the sustained enlargement of the penis for a longer period of time during sexual activity.

Contraindications

Avana tablets (Generic Avanafil tablets) are contraindicated in patients with a known hypersensitivity to Avanafil or any inactive ingredient present in Avana tablets.
